Ovalbumin Specific Immune Response in C57BL/6 Mice Immunized with Recombinant Mycobacterium smegmatis

Abstract
|
|
|
Construction of mycobacterial expression vector is the most critical for the development of live multivaccine vehicle with mycobacteria such as Bacillus Calmett-Guerin(BCG). In this study, a mycobacteri-E. Coli shuttle vector, pEYO was
constructed
for
manuplating DNA in E. coli and for expressing model antigen in mycobacteria. The expression vector pEYO carries mycobacterial autonomous heat shock protein 70(hsp70) promotor; kanamycin resistance gene of Tn903 in aph gene as a selection marker;
a
replication origin of M. fortuitum; also a replication origin of E. coli; and the gent for ovalbumin as model antigenic protein. M. smegmatis (mc2-155) was transformed with the plasmid pEYO by electroporation and the epression of recombinant
ovalbumin
by the transformants wee confirmed by western blot analysis. C57BL/6 mice immunized intraperitoneally with the recombinant M. smegmatis, induced the IgG1 isotype antibodies against the model antigenic protein ovablumin, successfully.
